Product Description

Step 1: Crystal Violet

Cover slide with Crystal Violet Reagent for one minute. Rinse slide with deionized or tap water.

Step 2: Grams Iodine

Cover slide with Iodine Reagent for one minute. Gently rinse the slide with deionized or tap water and allow to drain.

Step 3: Decolorizer

Tilt the slide and flood with a few drops of Decolorizer until no violet color runs off. This will usually take 10 seconds or less depending on the thickness of the specimen. Do not over decolorize. Rinse slide gently with deionized or tap water.

Step 4: Advanced Counter Stain

Cover slide with Advanced Counterstain for one minute. Rinse slide gently with deionized or tap water. The rinse water in this step should be slightly pink. Do not wash excessively. Allow slide to drain and air dry, or gently dry with lint-free bibulous paper or paper towel.

Weak staining is a leading cause of misidentification of bacteria. Gram-positives will retain a dark blue to violet, even those that are easily decolorized. Gram-negatives stain a vivid and distinct pink to red color. The combination provides a high degree of contrast for easy and reliable interpretation. When mixed on a slide, Gram-positives and negatives will be easier to distinguish due to an overall brighter stain.
